Tirzepatide is a 39-amino-acid synthetic peptide built on a modified GIP backbone, engineered to activate both the glucose-dependent insulinotropic polypeptide (GIP) receptor and the glucagon-like peptide-1 (GLP-1) receptor from a single sequence. A C20 fatty diacid moiety is conjugated to the lysine at position 20, which promotes reversible albumin binding and gives the molecule a markedly extended circulating half-life compared with native incretins. Research interest centres on what dual incretin engagement does that single-receptor agonism does not. Published in-vitro work characterises its biased signalling at the GLP-1 receptor — a comparatively weaker β-arrestin recruitment profile relative to cAMP accumulation — and its balanced potency across the two receptor populations. Preclinical models are used to examine insulinotropic response, gastric emptying kinetics, and adipose and hepatic substrate handling under combined GIP/GLP-1 signalling. Every batch is assayed by RP-HPLC for purity and confirmed by LC-MS for molecular identity, with the chromatograms and mass spectra published against the batch number.

Handling & reconstitution

Supplied lyophilised under vacuum. Allow the vial to reach room temperature before opening — introducing diluent into a cold vial draws condensation into the cake. Reconstitute with bacteriostatic water, directing the stream down the inner wall of the vial rather than onto the powder directly. The lipidated side chain makes tirzepatide prone to foaming and surface denaturation, so swirl gently until dissolved and never shake or vortex. Full dissolution typically takes one to two minutes; the solution should finish clear and colourless. Discard the vial if the reconstituted solution is cloudy, tinted, or shows visible particulates.

Storage

Lyophilised: stable at -20 °C in the sealed vial, protected from light and moisture. Short transit periods at ambient temperature do not compromise the lyophilisate. Reconstituted: 2–8 °C, used within approximately four weeks. Freezing an aqueous tirzepatide solution is not advised — the fatty acid chain aggregates across freeze-thaw cycles, and repeated cycling measurably degrades the peptide.
