Research summary

Thymulin is a nonapeptide of thymic epithelial origin with an unusual and defining requirement: it is biologically active only when complexed with a zinc ion. The zinc-free peptide is inactive, and the coupling is reversible, which means the availability of zinc directly governs the amount of active hormone present — a property that made thymulin a reference point in research linking trace element status to immune function. Published work describes it as acting on T cell differentiation and maturation, with studies examining effects on T cell markers, interleukin-2 production and natural killer cell activity. Circulating levels decline sharply with age in parallel with thymic involution, and separately fall in zinc deficiency, which is the basis for much of the immunosenescence literature involving it. A distinct strand of research examines analogues of thymulin in neuroinflammatory and nociceptive models. Each lot is assayed by RP-HPLC and confirmed by LC-MS, with the batch certificate published against the lot number.

Handling & reconstitution

Supplied lyophilised. Bring the vial to room temperature before opening. Reconstitute with bacteriostatic water introduced gently down the inner wall. The peptide itself dissolves readily, but the important handling point is the zinc dependency: avoid any buffer containing chelating agents, EDTA above all, which will strip the zinc and render the peptide biologically inactive while leaving it chromatographically intact. For work requiring the active zinc-bound form, the working buffer should supply zinc rather than sequester it. Swirl gently; do not vortex.

Storage

Lyophilised: -20 °C, sealed and dry, protected from light. Reconstituted: 2–8 °C, used within approximately two to three weeks. Because zinc binding is reversible, prolonged storage in solution — particularly in dilute form — allows progressive dissociation of the complex, so activity can decline while purity assays still look acceptable. Prepare working solutions close to the time of use, avoid chelator-containing vessels and buffers, and do not freeze-thaw repeatedly.
