SS-31, also known as elamipretide, is a tetrapeptide — D-Arg-Dmt-Lys-Phe-NH2 — belonging to the Szeto-Schiller class of mitochondria-targeted peptides. It incorporates a D-arginine residue and 2',6'-dimethyltyrosine, and its alternating aromatic-cationic structure is what drives its selective accumulation in the inner mitochondrial membrane. Its described mechanism is distinctive: rather than acting on a receptor, published work reports that SS-31 binds cardiolipin, the signature phospholipid of the inner mitochondrial membrane, and in doing so influences cristae architecture and the organisation of the electron transport chain supercomplexes that cardiolipin scaffolds. Research examines improved electron transport efficiency, reduced reactive oxygen species leak, and preservation of mitochondrial structure under stress. Importantly, its accumulation is described as independent of membrane potential, meaning it reaches depolarised and dysfunctional mitochondria that potential-driven targeting strategies do not. Handling & reconstitution
Supplied as a lyophilised powder. Allow the vial to warm to room temperature before opening. Reconstitute with bacteriostatic water added down the inner wall. SS-31 is strongly cationic and highly water-soluble, dissolving within seconds under gentle swirling. Its positive charge does cause some surface adsorption at low concentrations, so use low-binding tubes for dilute working solutions. Avoid buffers containing anionic detergents or lipids, which will bind the peptide and remove it from solution.
Storage
Lyophilised: -20 °C, sealed and dry, protected from light. Reconstituted: 2–8 °C, used within approximately four weeks. The D-amino acid and the dimethyltyrosine substitution make SS-31 resistant to peptidase degradation, and it is among the more stable compounds in the catalogue in solution. Protect from light — the dimethyltyrosine is the photosensitive element — and avoid repeated freeze-thaw cycling.
