Research summary
Semax is a synthetic heptapeptide constructed from the ACTH(4-10) fragment with a proline-glycine-proline tripeptide appended at the C-terminus. The ACTH fragment alone carries neurotropic activity but is degraded almost immediately by plasma peptidases; the Pro-Gly-Pro tail sterically protects the sequence and substantially extends its half-life. Critically, the fragment lacks the residues responsible for ACTH's corticotropic activity, so it does not stimulate adrenal steroidogenesis. Published research, much of it originating in Russian neuropharmacology, describes effects on brain-derived neurotrophic factor and nerve growth factor expression, modulation of the dopaminergic and serotonergic systems, and activity in cerebral ischaemia models. In-vitro work examines its influence on neuronal survival under hypoxic conditions and on trophic factor signalling cascades. Purity is determined by RP-HPLC and identity confirmed by LC-MS, with batch-linked analytical data published for every lot released.
Handling & reconstitution
Supplied lyophilised. Bring the vial to room temperature before opening. Reconstitute with bacteriostatic water added down the inner wall. Semax dissolves readily with gentle swirling. Note the methionine residue in the sequence — it is the most oxidation-prone amino acid, so minimise the time the open vial spends exposed to air and avoid introducing oxidising agents. The solution should be clear and colourless.
Storage
Lyophilised: -20 °C, sealed and dry, protected from light. Reconstituted: 2–8 °C, used within approximately two to three weeks. Methionine oxidation is the limiting degradation pathway and it proceeds slowly even under refrigeration, so the unmodified peptide has a shorter usable window than its acetylated counterpart. Keep the vial full and closed rather than leaving significant headspace, and do not freeze-thaw repeatedly.
