Research summary
PT-141, also known as bremelanotide, is a cyclic heptapeptide and the principal active metabolite of Melanotan II, formed by deletion of the C-terminal amide. That single structural difference substantially changes its receptor profile. Where Melanotan II is broadly non-selective, PT-141 shows preferential activity at MC4R and MC3R with markedly reduced MC1R agonism — meaning it largely lacks the melanogenic activity of its parent compound. That separation is what made it a distinct research subject rather than a metabolite of one. Published work examines MC4R signalling in the central nervous system, where the receptor is implicated in energy homeostasis and in the regulation of sexual response pathways; its mechanism is described as central and neurally mediated rather than acting on peripheral vascular tissue, which distinguishes it mechanistically from the phosphodiesterase inhibitor class. Handling & reconstitution
Supplied lyophilised. Allow the vial to reach room temperature before opening. Reconstitute with bacteriostatic water added gently down the vial wall. The cyclic structure makes PT-141 comparatively robust and it dissolves readily with swirling. Tryptophan in the sequence makes it light-sensitive; keep it out of direct illumination during preparation and storage.
Storage
Lyophilised: -20 °C, sealed, dry and protected from light. Reconstituted: 2–8 °C, used within approximately four weeks, kept dark. Like Melanotan II, the cyclic backbone gives good solution stability and photo-oxidation of the tryptophan residue is the limiting factor rather than hydrolysis or aggregation. Store wrapped or in amber glass and avoid freeze-thaw cycling.
