Research summary
P21 is a synthetic peptide derived from a biologically active region of ciliary neurotrophic factor, developed to reproduce the neurogenic activity of the parent protein in a small, more tractable molecule. CNTF itself is a 200-residue cytokine that does not cross the blood-brain barrier and has a difficult profile as a research tool; the peptide was constructed to isolate the activity of interest from that context. Published in-vitro work describes stimulation of neurite outgrowth and enhanced survival in cultured neurons, and preclinical studies examine hippocampal neurogenesis and synaptic protein expression. A distinct strand of the literature reports effects on tau phosphorylation and amyloid processing in transgenic models, which is the basis for its appearance in neurodegeneration research. It is a comparatively recent compound and the body of published work remains small relative to the older nootropic peptides. Every lot is assayed by RP-HPLC and verified by LC-MS, with a batch-linked Certificate of Analysis published for each release.
Handling & reconstitution
Supplied lyophilised. Bring the vial to room temperature before opening. Reconstitute with bacteriostatic water added slowly against the inner wall. P21 is moderately soluble and generally dissolves with gentle swirling over one to two minutes; if material remains after that, allow it to stand rather than escalating to agitation or heat. The solution should finish clear. Discard if haze or particulate persists.
Storage
Lyophilised: -20 °C, sealed and dry, protected from light. Reconstituted: 2–8 °C, used within approximately two to three weeks. As a longer synthetic peptide with defined secondary structure, P21 is less stable in solution than the short nootropic peptides here. For work extending beyond that window, aliquot into single-use volumes and freeze once at -20 °C rather than cycling a single vial.
