Research summary
N-Acetyl Semax Amidate is a doubly modified form of Semax: the N-terminus is acetylated and the C-terminus amidated. Neither change alters the core ACTH(4-10)-Pro-Gly-Pro sequence, and both target the same problem — terminal degradation by exopeptidases. Blocking both termini removes the free amino and carboxyl groups that aminopeptidases and carboxypeptidases recognise, and published work on terminally modified peptides describes markedly increased metabolic stability as a result. The amidation also removes the C-terminal negative charge, which increases lipophilicity and is the basis for the interest in this variant over unmodified Semax. It is studied in the same contexts — neurotrophic factor expression, monoaminergic modulation, neuroprotection under ischaemic stress — with the modifications treated as a pharmacokinetic rather than pharmacodynamic variable. Each lot is assayed by RP-HPLC for purity and LC-MS for identity, with the certificate searchable against the vial's batch number.
Handling & reconstitution
Supplied as a lyophilised powder. Equilibrate to room temperature before reconstituting. Add bacteriostatic water slowly against the vial wall and swirl gently until dissolved. The terminal modifications increase lipophilicity slightly, so dissolution can take marginally longer than unmodified Semax — allow a minute rather than forcing it with agitation. Expect a clear, colourless solution.
Storage
Lyophilised: -20 °C, sealed, dry and protected from light. Reconstituted: 2–8 °C, used within approximately four weeks — a longer window than unmodified Semax, since the blocked termini remove the exopeptidase-mediated degradation route. The methionine residue remains oxidation-sensitive, however, so light protection and minimal air exposure still apply. Avoid freeze-thaw cycling.
