Research summary

MOTS-c is a 16-amino-acid mitochondrial-derived peptide encoded not in nuclear DNA but within the mitochondrial 12S rRNA gene — one of a small group of peptides that established mitochondria as a source of signalling molecules rather than solely a site of energy production. Its discovery is comparatively recent, and much of the literature is still mapping basic biology. Published research places it in the folate-methionine cycle, where it appears to influence purine biosynthesis and, through accumulation of the intermediate AICAR, to activate AMP-activated protein kinase. Studies also describe its translocation to the nucleus under metabolic stress, where it associates with stress-response transcription factors — an unusual property for a mitochondrially encoded peptide. In-vitro and preclinical models are used to examine insulin sensitivity, exercise-response signalling and age-associated changes in circulating levels. Each lot is verified by RP-HPLC for purity and LC-MS for identity, with the certificate tied to the batch number on the vial.

Handling & reconstitution

Supplied lyophilised. Equilibrate to room temperature before opening — this one is worth doing carefully, as the cake is light and easily disturbed. Reconstitute with bacteriostatic water added slowly down the vial wall. MOTS-c is short and dissolves quickly with gentle swirling. Do not sonicate or warm the vial to speed dissolution. The reconstituted solution should be clear and colourless.

Storage

Lyophilised: -20 °C, sealed and dry. Stable in the dry state and tolerant of shipping at ambient temperature. Reconstituted: 2–8 °C, used within approximately three to four weeks. For longer-term work, aliquot into single-use volumes and freeze once at -20 °C rather than repeatedly cycling a single vial — MOTS-c tolerates one freeze considerably better than several.
