Research summary
Melanotan II is a cyclic lactam analogue of alpha-melanocyte-stimulating hormone, in which a covalent bridge between residues constrains the peptide into the conformation that binds the melanocortin receptors. That cyclisation is what gives it both its potency and its resistance to enzymatic degradation relative to the linear parent hormone. Its research value and its research limitation are the same property: it is a non-selective agonist across the melanocortin receptor family, engaging MC1R, MC3R, MC4R and MC5R. Published in-vitro work uses it as a broad-spectrum reference agonist when characterising receptor binding affinities and downstream cAMP signalling, and preclinical studies examine MC1R-mediated melanogenesis alongside the MC3R and MC4R contributions to energy homeostasis signalling. Its lack of selectivity is precisely why more selective analogues were subsequently developed from it. Purity is determined by RP-HPLC and identity confirmed by LC-MS, with batch-linked chromatograms published for every lot released.
Handling & reconstitution
Supplied lyophilised. Bring the vial to room temperature before opening. Reconstitute with bacteriostatic water added down the inner wall and swirl gently. The cyclic structure makes Melanotan II more robust than a comparable linear peptide, and it dissolves readily within a minute. It contains tryptophan and is light-sensitive as a result — keep the vial shielded from direct illumination during handling rather than working under a bright bench lamp.
Storage
Lyophilised: -20 °C, sealed, dry and protected from light. Reconstituted: 2–8 °C, used within approximately four weeks, stored in the dark. The lactam bridge confers good stability in solution — better than most linear peptides here — but tryptophan photo-oxidation remains the principal degradation route and it proceeds under ordinary room lighting. Store the reconstituted vial wrapped or in amber glass, and avoid freeze-thaw cycling.
