Research summary

Melanotan I, also known as afamelanotide, is a linear 13-amino-acid analogue of alpha-MSH differing from the native hormone by two substitutions — a norleucine at position 4 and a D-phenylalanine at position 7. Both changes confer resistance to enzymatic degradation while retaining the linear architecture of the parent hormone. The contrast with Melanotan II is the reason both are catalogued. Melanotan I is substantially more selective for MC1R, the receptor expressed on melanocytes that governs eumelanin synthesis, and shows considerably less activity at MC3R and MC4R. That selectivity makes it the cleaner tool compound for research isolating MC1R signalling — melanogenesis, tyrosinase induction, and the photoprotective response — from the broader melanocortin effects that confound work with the non-selective cyclic analogue. It is also the better-characterised of the two in formal clinical literature. Each lot is assayed by RP-HPLC and verified by LC-MS, with a batch-specific certificate published for every release.

Handling & reconstitution

Supplied as a lyophilised powder. Equilibrate the vial to room temperature before opening. Reconstitute with bacteriostatic water introduced slowly against the inner wall. As a linear 13-residue peptide, Melanotan I is more susceptible to mechanical stress than the cyclic analogue — swirl gently and allow a minute or two, and do not shake or vortex. It contains tryptophan, so protect it from direct light while working.

Storage

Lyophilised: -20 °C, sealed, dry and dark. Reconstituted: 2–8 °C, used within approximately two to three weeks — a shorter window than Melanotan II, since the linear structure lacks the conformational stabilisation the lactam bridge provides. Store protected from light throughout and do not freeze-thaw repeatedly.
