Research summary

LL-37 is the 37-amino-acid C-terminal fragment of human cathelicidin antimicrobial protein hCAP18, released by proteinase-3 cleavage of the precursor. It is the only cathelicidin found in humans, and it is amphipathic and strongly cationic — properties that define both its biology and its awkwardness at the bench. Its antimicrobial activity is membrane-disruptive rather than target-specific: the cationic helix associates with anionic bacterial membranes and compromises their integrity, a mechanism that gives broad-spectrum activity and makes resistance development slow. Equally significant in the literature is its immunomodulatory role — published work describes chemotactic activity for neutrophils, monocytes and T cells, neutralisation of lipopolysaccharide, and involvement in wound repair and angiogenesis. It is one of the most extensively studied host defence peptides. Purity by RP-HPLC and identity by LC-MS, with batch-linked chromatograms published for every lot.

Handling & reconstitution

LL-37 is the most handling-sensitive compound in the catalogue and needs care that the shorter peptides do not. Its strong positive charge causes it to adsorb readily onto glass and standard polypropylene surfaces, which can remove a substantial fraction of a dilute preparation from solution before the experiment begins. Use low-binding tubes and tips throughout, and prepare dilute working solutions immediately before use rather than storing them. Reconstitute with sterile water or dilute acetic acid rather than saline — high ionic strength buffers reduce solubility and can trigger precipitation. Add the diluent slowly, swirl gently, and do not vortex: the amphipathic helix aggregates readily under mechanical stress.

Storage

Lyophilised: -20 °C or below, sealed and dry. Keep it in the dry state until genuinely needed — this is the only form in which LL-37 is reliably stable. Reconstituted: prepare a concentrated stock, aliquot immediately into single-use low-binding vials, and freeze at -20 °C or below. Freeze-thaw cycling causes irreversible aggregation, so thaw each aliquot once and discard the remainder. Refrigerated stock solutions should be used within days rather than weeks.
