Research summary
Liraglutide is a GLP-1 analogue sharing 97% sequence homology with native human GLP-1(7-37). It differs by a single lysine-to-arginine substitution at position 34 and a C16 palmitic acid chain attached via a glutamic acid spacer to the lysine at position 26. That acylation drives albumin binding and self-association into heptameric assemblies at the injection site, which together slow absorption and clearance relative to the native hormone. It is the longest-established acylated GLP-1 analogue and consequently serves as a reference standard in comparative incretin research. In-vitro studies use it to benchmark receptor binding affinity, cAMP signalling and receptor trafficking against newer analogues. Its shorter circulating half-life relative to semaglutide makes it useful where a more dynamic pharmacokinetic profile is wanted in a model. Each lot is verified by RP-HPLC for purity and LC-MS for identity, with the certificate tied to the batch number printed on the vial.
Handling & reconstitution
Supplied as a lyophilised powder. Bring to room temperature before reconstituting. Add bacteriostatic water gently against the inner wall. Liraglutide self-associates in solution, which is expected behaviour and not a sign of degradation — but it does mean the vial should be swirled rather than shaken, and given time to clarify. Do not vortex. The solution should be clear once fully dissolved.
Storage
Lyophilised: -20 °C, sealed, protected from light. Reconstituted: 2–8 °C, used within approximately four weeks. Liraglutide is comparatively tolerant of refrigerated storage but should not be frozen in solution, and should be kept out of direct light throughout.
