Larazotide, also catalogued as AT-1001, is a synthetic octapeptide developed as a tight junction regulator. It was derived from a Vibrio cholerae protein and acts as an antagonist of zonulin, the endogenous modulator of intestinal epithelial tight junction permeability. Its mechanism distinguishes it from most compounds in this catalogue: it acts locally at the luminal surface of the intestinal epithelium rather than systemically, and published work describes it as promoting the reassembly of tight junction complexes and reducing paracellular permeability rather than acting on a signalling receptor in the conventional sense. Research models concentrate on barrier integrity — in-vitro work uses transepithelial electrical resistance and macromolecule flux across epithelial monolayers as primary endpoints, with gliadin-induced permeability the most frequently modelled challenge. Handling & reconstitution

Supplied as a lyophilised powder. Equilibrate to room temperature before opening. Reconstitute with sterile or bacteriostatic water added slowly against the vial wall. Larazotide is a short peptide and dissolves readily with gentle swirling, though its glycine-rich sequence makes it somewhat less soluble than the smallest tripeptides here — allow a minute or two and do not force it with agitation. The solution should finish clear.

Storage

Lyophilised: -20 °C, sealed and dry. Reconstituted: 2–8 °C, used within approximately three to four weeks. Larazotide is stable in neutral aqueous solution but its activity is pH-dependent in epithelial models, so any working buffer should be pH-controlled and prepared fresh. Avoid freeze-thaw cycling; aliquot before freezing if longer storage is needed.
