Research summary
Humanin is a 24-amino-acid peptide encoded within the mitochondrial 16S ribosomal RNA gene — the first mitochondrial-derived peptide identified, and the discovery that established mitochondrial DNA as a source of signalling peptides rather than solely respiratory chain components. It was found in a screen of surviving neurons from Alzheimer's-affected brain tissue. Published research describes cytoprotective activity across a range of stress models, with mechanistic work reporting interaction with pro-apoptotic Bcl-2 family proteins including Bax, and signalling through a trimeric receptor complex involving CNTFR, WSX-1 and gp130 that activates STAT3. Studies also examine its interaction with IGF-binding protein-3 and its relationship to metabolic regulation and insulin sensitivity. Circulating levels are reported to decline with age, which links it to the broader mitochondrial-derived peptide literature alongside MOTS-c. Handling & reconstitution
Supplied lyophilised. Bring the vial to room temperature before opening. Reconstitute with bacteriostatic water added slowly down the vial wall. Humanin is a 24-residue peptide with a hydrophobic segment and is less freely soluble than the short mitochondrial peptides — allow two to three minutes with gentle swirling for complete dissolution, and resist the temptation to vortex, which promotes aggregation of the hydrophobic region. The solution should finish clear; discard if haze persists after standing.
Storage
Lyophilised: -20 °C, sealed, dry and protected from light. Reconstituted: 2–8 °C, used within approximately two to three weeks. Humanin is more aggregation-prone in solution than MOTS-c owing to its length and hydrophobic content, and it also contains a methionine residue susceptible to oxidation. Aliquot into single-use volumes and freeze once if the vial will not be consumed within that window; inspect for haze before each use.
