Research summary
Glutathione is a tripeptide — γ-glutamyl-cysteinyl-glycine — and the principal low-molecular-weight antioxidant of the mammalian cell, present at millimolar concentrations in most tissues. Its structure is unusual: the glutamate is linked through its side-chain γ-carboxyl rather than the α-carboxyl, an arrangement that makes it resistant to ordinary aminopeptidases. The cysteine thiol is the functional group and the reason for the research interest. Glutathione cycles between the reduced form (GSH) and the oxidised disulfide dimer (GSSG), and the ratio between them is the standard measure of cellular redox state. Published work covers its role as substrate for glutathione peroxidases in peroxide detoxification, for glutathione S-transferases in xenobiotic conjugation, and in protein S-glutathionylation as a redox signalling mechanism. It is supplied here in the reduced form. Handling & reconstitution
Glutathione is chemically the most reactive compound in this catalogue and its handling reflects that. The free thiol oxidises readily on contact with atmospheric oxygen, converting reduced GSH to the oxidised GSSG dimer — and since almost all research applications depend on the reduced form, that conversion is a direct loss of the material's usefulness. Minimise air exposure: open the vial only when ready, reconstitute promptly, and reseal immediately. Reconstitute with sterile water. Avoid alkaline buffers, which accelerate thiol oxidation sharply, and avoid contact with metal ions such as copper and iron, which catalyse it. Prepare working solutions immediately before use rather than in advance.
Storage
Lyophilised: -20 °C, tightly sealed, dry and protected from light. The dry reduced form is stable; leave it sealed until needed. Reconstituted: this is where glutathione differs most sharply from the peptides here. Oxidation in aqueous solution proceeds within hours to days rather than weeks, and refrigeration slows it without stopping it. Prepare fresh and use the same day wherever possible. If a solution must be held, aliquot into single-use volumes with minimal headspace, freeze immediately at -20 °C or below, and thaw once. A solution that has developed a yellow tint or a sulfurous odour should be discarded.
