Research summary

GHK-Cu is a copper-binding tripeptide — glycyl-L-histidyl-L-lysine — complexed with a copper(II) ion. The free peptide occurs naturally in human plasma, where its concentration declines markedly with age, and it has an exceptionally high affinity for Cu(II); the complex, not the bare peptide, is the biologically active species in most of the published work. Research centres on its role as a copper delivery vehicle and signalling molecule. Copper is an obligatory cofactor for lysyl oxidase, the enzyme that cross-links collagen and elastin, which places GHK-Cu directly in the extracellular matrix remodelling literature. In-vitro studies describe stimulation of collagen and glycosaminoglycan synthesis in fibroblast culture, modulation of matrix metalloproteinase and TIMP expression, and antioxidant activity. Gene-expression work reporting broad transcriptional effects has extended interest well beyond dermal models. Purity is confirmed by RP-HPLC and identity by LC-MS, with batch-specific analytical data published for every lot.

Handling & reconstitution

Supplied lyophilised. The powder is blue — this is the copper complex and is expected, not a contaminant. Allow the vial to reach room temperature before opening. Reconstitute with bacteriostatic water. The solution will be distinctly blue once dissolved; loss of that colour indicates the copper has dissociated and the complex has broken down. Avoid any buffer containing chelating agents — EDTA in particular will strip the copper from the peptide and inactivate it. Avoid contact with reducing agents, which reduce Cu(II) to Cu(I) and destabilise the complex. Swirl gently; do not vortex.

Storage

Lyophilised: -20 °C, sealed and dry, protected from light. The dry complex is stable and retains its blue colour indefinitely under these conditions. Reconstituted: 2–8 °C, used within approximately four weeks, protected from light. Watch the colour as a stability indicator — a solution that has faded, greened or thrown a precipitate has lost complex integrity and should be discarded regardless of how recently it was prepared. Do not store in metal-contacting containers.
