Research summary

FOXO4-DRI is a D-retro-inverso peptide: the sequence is reversed and built entirely from D-amino acids, an arrangement that preserves the spatial arrangement of side chains while making the molecule essentially invisible to proteases. It is derived from the FOXO4 transcription factor's p53-interacting domain. Its research application is senolytic and the mechanism is specific. In senescent cells, FOXO4 sequesters p53 in the nucleus, preventing its mitochondrial translocation and thereby blocking the apoptotic programme that would otherwise remove the cell. The peptide competes for that interaction, displacing p53 and permitting apoptosis to proceed — selectively in senescent cells, since the FOXO4-p53 interaction is characteristic of the senescent state. Published in-vitro work examines selective clearance of senescent cells in culture, with preclinical models addressing markers of tissue ageing. Purity is confirmed by RP-HPLC and identity by LC-MS, with batch-specific chromatograms published for every lot released.

Handling & reconstitution

FOXO4-DRI is one of the more difficult compounds here to get into solution and should not be handled like a short hydrophilic peptide. It is a comparatively long sequence with substantial hydrophobic content and dissolves poorly in water alone. Common practice is to prepare a concentrated stock in a small volume of DMSO, then dilute into the aqueous working buffer. Add the aqueous phase to the organic stock slowly with mixing and watch the point of dilution for precipitation. Do not vortex or sonicate aggressively. If material remains after several minutes of gentle swirling, the diluent is the problem, not the mixing.

Storage

Lyophilised: -20 °C or below, sealed and dry, protected from light. DMSO stock: aliquot into single-use volumes and store at -20 °C. DMSO absorbs atmospheric moisture on each opening, which lowers the effective solvent quality and eventually causes the peptide to precipitate out of stock — minimise open-close cycles and seal tightly. The D-amino acid backbone makes the peptide resistant to enzymatic degradation, so protease contamination is not the concern it would be for an L-peptide; aggregation and freeze-thaw damage are. Aqueous working dilutions should be made fresh.
