Extensive modification — including N-terminal hexanoic acid acylation and C-terminal benzylamide capping — makes it far more lipophilic and metabolically stable than the peptide it originates from. Its described mechanism is unusual and is the reason for the research interest. Published work characterises it not as a receptor agonist in the conventional sense but as a positive modulator of hepatocyte growth factor signalling at its receptor c-Met, acting to potentiate HGF activity rather than to substitute for it. In-vitro studies examine dendritic spine formation and synaptogenesis in hippocampal neuron culture, with reported potency in these assays substantially exceeding that of BDNF in the same models. Handling & reconstitution

Dihexa does not handle like the water-soluble peptides in this catalogue and should not be treated as one. Its lipophilicity — the property that gives it blood-brain-barrier penetration — makes it poorly soluble in water. It is conventionally dissolved first in a small volume of an organic solvent such as DMSO to produce a concentrated stock, which is then diluted into the aqueous working medium. Attempting to reconstitute it directly in bacteriostatic water will generally leave undissolved material. Add the aqueous diluent to the organic stock slowly and with mixing, and watch for precipitation at the point of dilution; if the compound crashes out, the stock concentration is too high for the intended final volume.

Storage

Dry compound: -20 °C, sealed and protected from moisture and light. It is stable in this form. Organic stock solution: aliquot into single-use volumes and store at -20 °C. DMSO is strongly hygroscopic and absorbs atmospheric water on every opening, which progressively degrades stock concentration and eventually causes the compound to precipitate — sealing tightly and minimising the number of freeze-thaw and open-close cycles matters more here than the storage temperature itself. Aqueous working dilutions should be prepared fresh and not stored.
