Research summary
CJC-1295 with DAC is the same modified GHRH(1-29) analogue as the non-DAC form, with one substantial addition: a maleimidopropionic acid group — the Drug Affinity Complex — conjugated to the C-terminal lysine. That linker forms a covalent bond with cysteine-34 of circulating serum albumin, tethering the peptide to a long-lived carrier protein and extending its half-life from minutes to days. The two variants are studied as a matched pair, and the comparison is the interesting part. The non-DAC form produces a discrete pulse of GHRH receptor stimulation; the DAC form produces a sustained elevation in circulating peptide, which published work describes as raising the baseline of the secretion profile rather than reproducing native pulsatility. Research using it therefore addresses a different question — what continuous GHRH receptor occupancy does to somatotroph signalling and downstream feedback. Purity by RP-HPLC, identity by LC-MS, with batch-specific chromatograms published for every lot released.
Handling & reconstitution
Supplied lyophilised. Equilibrate the sealed vial to room temperature before opening. Reconstitute with bacteriostatic water introduced down the inner wall. The DAC linker carries a reactive maleimide group, which is what makes the albumin conjugation work — it also means the peptide should be handled without unnecessary delay once in solution, and should not be exposed to thiol-containing buffers, which will compete for the linker. Swirl gently until clear. Do not vortex.
Storage
Lyophilised: -20 °C, sealed and dry, protected from light. Keeping the vial genuinely dry matters more here than for most compounds — the maleimide group hydrolyses on exposure to moisture, and a hydrolysed linker will no longer conjugate. Reconstituted: 2–8 °C, used within approximately three to four weeks. Avoid freeze-thaw cycling and avoid any buffer containing free thiols.
