CJC-1295 without DAC — also catalogued as modified GRF(1-29) — is a 29-amino-acid analogue of the biologically active N-terminal fragment of growth hormone-releasing hormone, carrying four amino acid substitutions relative to the native sequence. Those substitutions target the enzymatic and chemical liabilities of native GHRH: resistance to dipeptidyl peptidase-4 cleavage, reduced asparagine deamidation, and suppressed methionine oxidation. Critically, it lacks the Drug Affinity Complex present in the DAC variant, so it does not bind albumin and its half-life is measured in minutes rather than days. That short window is a feature in research contexts, not a limitation — it produces a discrete, pulse-like GHRH receptor stimulus, which is what studies of episodic secretion require. It is frequently examined alongside GHS-R1a agonists to characterise the synergy between the two receptor pathways. Each lot is assayed by RP-HPLC and verified by LC-MS, with the certificate searchable against the vial's batch number.
Handling & reconstitution
Supplied lyophilised. Allow the vial to warm to room temperature before reconstituting. Add bacteriostatic water slowly against the vial wall. The 29-residue chain is more susceptible to shear than the short secretagogue peptides — swirl gently until dissolved and do not shake or vortex. Discard if the solution does not finish clear.
Storage
Lyophilised: -20 °C, sealed, protected from light and moisture. The oxidation-resistant substitutions improve dry-state shelf life relative to native GHRH but do not eliminate the need for cold storage. Reconstituted: 2–8 °C, used within approximately two to three weeks. This is a shorter window than the DAC variant — without albumin binding the peptide is more exposed in solution, and it degrades faster. Protect from light and do not freeze once reconstituted.
