Research summary

Cagrilintide is a long-acting amylin analogue engineered for stability where native human amylin is not. Human amylin aggregates readily into amyloid fibrils, which makes it impractical as a research tool; cagrilintide's sequence borrows stabilising features associated with non-amyloidogenic amylins and adds a fatty-acid modification for albumin binding and extended half-life. It is a non-selective agonist across the amylin receptor family and the calcitonin receptor, and research interest lies largely in that breadth. Published in-vitro work characterises its binding profile across calcitonin receptor and RAMP-modified amylin receptor subtypes; preclinical models examine amylin signalling in the area postrema and its interaction with concurrent GLP-1 receptor activation, a combination that has become a significant line of investigation in metabolic pharmacology. Purity is established by RP-HPLC and identity by LC-MS, with batch-linked chromatograms published for every lot.

Handling & reconstitution

Supplied lyophilised. Allow the vial to warm to room temperature before opening. Reconstitute with bacteriostatic water introduced slowly down the vial wall. Amylin-family peptides are aggregation-prone by nature, and although cagrilintide is engineered against this, mechanical agitation still encourages it — swirl gently, never shake, and allow the solution to clarify on its own. Inspect against a light source before use. Any cloudiness or fibrous material indicates aggregation, and the vial should be discarded.

Storage

Lyophilised: -20 °C, sealed and dry. The lyophilisate is the stable form and should be kept that way until needed. Reconstituted: 2–8 °C, used within approximately three to four weeks — a shorter window than the GLP-1 analogues, because amylin analogues aggregate more readily in solution over time. Do not freeze once reconstituted, and inspect for haze before each use.
