Research summary

Argireline is acetyl hexapeptide-8, a six-residue peptide derived from the N-terminal domain of SNAP-25 and acetylated at the N-terminus for stability. It is the original and most extensively studied member of the SNARE-competing peptide class, and SNAP-8 was subsequently developed from it. Its described mechanism is competitive inhibition of SNARE complex formation. Vesicle fusion and neurotransmitter release at the neuromuscular junction require SNAP-25 to assemble with syntaxin and VAMP into a ternary complex; a peptide reproducing the relevant SNAP-25 segment can occupy that position and destabilise the assembly. Published in-vitro work uses chromaffin cell models to measure catecholamine release under peptide exposure, and dermal formulation studies examine penetration and matrix effects. It is the benchmark compound against which newer peptides in this class are compared. Purity is determined by RP-HPLC and identity by LC-MS, with batch-linked chromatograms published for every lot released.

Handling & reconstitution

Supplied lyophilised. Bring the vial to room temperature before opening. Reconstitute with sterile or bacteriostatic water added slowly down the inner wall. Argireline is strongly hydrophilic and among the easiest compounds here to get into solution — it dissolves essentially on contact with gentle swirling, with no co-solvent required. In formulation work it is stable across a broad pH range but performs best in neutral to slightly acidic conditions; add it late in the process and avoid high-temperature processing steps.

Storage

Lyophilised: -20 °C, sealed and dry. The acetylated N-terminus blocks aminopeptidase attack and the compound is very stable in the dry state. Reconstituted: 2–8 °C, used within approximately four to six weeks. With no oxidation-sensitive residues and a blocked N-terminus, Argireline is one of the more stable peptides in the catalogue in aqueous solution — microbial growth is the limiting factor rather than chemical degradation, so sterile technique on every withdrawal is what protects it. Avoid freeze-thaw cycling.
