5-Amino-1MQ is a small-molecule inhibitor rather than a peptide — a quinolinium compound included in this catalogue as a companion to the metabolic and mitochondrial research compounds. It is a selective, cell-permeable inhibitor of nicotinamide N-methyltransferase. NNMT catalyses the methylation of nicotinamide to 1-methylnicotinamide, consuming S-adenosylmethionine in the process. That reaction sits at a branch point: nicotinamide diverted to methylation is nicotinamide unavailable for salvage back into NAD+. Published research therefore examines NNMT inhibition as a route to influencing cellular NAD+ availability and methyl donor balance simultaneously. NNMT is highly expressed in adipose tissue and its expression is reported to be elevated in obesity models, which is the basis for the in-vitro and preclinical work on adipocyte metabolism, lipogenesis and energy expenditure.
